METHOTREXATE ACTION MECHANISM
Methotrexate acts through two different mechanisms for cancer and rheumatoid arthritis treatment. In cancer treatment, methotrexate allosterically inhibits dihydrofolate reductase, thereby specifically acting during DNA and RNA synthesis. While in rheumatoid arthritis treatment, Methotrexate inhibits enzymes of purine metabolism, accumulating adenosine or inhibiting T cell activation.
METHOTREXATE DOSAGE
Methotrexate is an oral medication and its dosage depends on your condition and how your body responds to medication. Methotrexate is usually recommended once or twice a week and not on a daily basis.

Common Side effects of Methotrexate include dizziness, drowsiness, loss of appetite and nausea.
While serious side effects of Methotrexate that require urgent medical help include diarrhea, difficulty in breathing, mouth and throat ulcers, skin rash, peeling of skin, weakness or tiredness, fainting spells, vomiting, yellow coloring of skin or eyes, sore throat, vision changes, black or tarry stools and blood in urine.
